Accuracy, Verification, and Application of Monitoring Spawning Grounds of Plecoglossus Altivelis Using Environmental Dna Analysis

Abstract: In recent years, the abundance of Ayu (Plecoglossus altivelis) has been decreasing in Japan. Therefore, it is necessary to assess the current abundance and to identify the factors leading to the decline of this fish species. In this study, we assessed the potential use of environmental DNA (eDNA) for monitoring P. altivelis in the spawning grounds. We employed eDNA monitoring in an artificial spawning ground in the Nahari River and applied this method to the Takatsu River to determine downstream migration and to identify suitable spawning habitats for P. altivelis. The results of the eDNA analysis from the Nahari River showed that e DNA can be useful for spawning-ground monitoring of P. altivelis and the results from the Takatsu River confirmed downstream migration of P. altivelis.
