Abstract: We examined the introgression of the stream caddisfly Stenopysche maramorata (Stenopsychidae, Trichoptera) in 4 catchments in Miyagi, Japan based on both nuclear DNA genotypes and mitochondrial DNA haplotypes. S. marmorata was widespread and was one of dominant species in riffle in Japanese river. At first, 8 microsatellites were employed genotyping for nucleic DNA (nDNA). A model-based Bayesian clustering analysis showed 2populations separated into upland and lowland populations. We then sequenced cytochrome c oxidase subunit I (COI) mitochondrial DNA (mtDNA). A phylogenetic gene tree also showed distinct 2 populations isolated by elevation. The majority individuals showed congruence of genetic markers and were classified into upland or lowland populations by both nDNA and mtDNA. A small proportion (ca. 8%) were hybrids, with incongruent hich have disagreed genetic patterns, occurred broadly. We found two types of hybrids (i. e. upland nDNA with lowland mtDNA and lowland nDNA with upland mtDNA). It indicated that gene flow between the populations was occurred both direction (i. e. from upland to lowland, from lowland to upland).
